Need more details to help:

PCIe card with SATA ports?

PCIe card for M.2 SATA card?

PCIe card for M.2 NVMe card?

Do you have a picture of something similar?

You can't boot from a M.2 NVMe PCIe drive in a Gen8 as it does not have a UEFI BIOS. If you use a M.2 SATA drive in this adapter you would need a SATA port to connect to it and from your other posts I think you are using the 5th SATA port for a DVD drive.

I have a similar card and as Scoondoggy suggests I have a M.2 drive connected to the 5th SATA port. I also have a NVME installed in the 2nd slot and although it cannot boot it still shows up as an additional drive

I'm using ASUS Hyper M.2 x4 Mini for data storage purposes and now waiting for Gigabyte CMT4032.
